This increase in glutamate sensitivity of the TNC can be further amplified by acetylcholine (ACh) and ketamine, both glutamate antagonists. This promotes neuronal firing which in turn reduces migraine aura intensity.

Ketamine also has an anticonvulsant effect, which may help to reduce the frequency of migraine headaches. This is because ketamine blocks the release of vasoactive peptides from the trigeminal system and causes constriction of cranial blood vessels.

Combining tryptophan and nicotinamide can be effective for treating ocular migraines. These amino acids may decrease headache frequency as well as alleviate glare- and light-induced pain in those suffering from migraines.
